Lafayette County was established on December 23, 1856, carved from Madison County and originally encompassing land that today also makes up neighboring Dixie County. It was named for the Marquis de Lafayette, the French general celebrated for his service alongside the Continental Army during the American Revolution.

Lafayette's popularity surged following his celebrated tour of the United States in the early 1820s, and Florida settlers of that era commemorated him by attaching his name to the new county decades later.

New Troy initially served as county seat until its courthouse burned down in 1892, prompting the seat to move to Mayo in 1893, where it has remained ever since. Farming and timber shaped the county's small, rural economy through the century that followed.
