Gadsden County was created on June 24, 1823 from Jackson County, becoming Florida's fifth county and part of what was then called Middle Florida. It was named for James Gadsden, an aide to territorial governor Andrew Jackson who later became a diplomat known for negotiating the 1854 Gadsden Purchase.

At its creation the county's boundaries were vast, stretching from the Suwannee River in the east to the Apalachicola River in the west, before later subdivisions reduced it to its present, more compact size.

Shade-grown tobacco became Gadsden County's signature crop by the late nineteenth century, cultivated under cheesecloth tents to produce wrapper leaf for cigars, and the industry dominated the local economy for decades before eventually fading in the twentieth century.
